A discontinuous buffer system for paper electrophoresis of human hemoglobins.

چکیده
NEW BUFFER SYSTEM has recently been introduced for the separation of proteins (1) and hemoglobins (2) on starch gel. It differs from the conventional barbital system most frequently used heretofore in that the buffer used for the supporting medium differs from that used in the buffer vessels. For this reason the buffer system is referred to as discontinuous. The discontinuous buffer system appears to be superior not only for electrophoretic separations using starch gel as a supporting medium (2) but for electrophoresis on paper as well. A method for paper electrophoresis of hemoglobins will be described, in which Tris-EDTA-borate buffer is used for the immersion of the paper and barbital buffer is used in the buffer vessels.

An analysis of some hemoglobin and haptoglobin phenotypes in Blantyre, Malawi.
Samples of blood and sera were obtained from unrelated adolescent and adult Bantus who were clinical patients at the Central Hospital. Males and females were pooled together for analysis. Hemolysates were analyzed by starch-gel electrophoresis methods using Tris-EDTA-borate buffer at pH 8.6 [1] and by cellulose-acetate electrophoresis in the same basic buffer system. Benzidine was used for stai...
متن کاملA new method for starch gel electrophoresis of human hemoglobins, with special reference to the determination of hemoglobin A2.
ONSIDERABLE EVIDENCE IS now available indicating that normal adult hemoglobin (hemoglobin A) is heterogeneous. Sharp and well-defined fractions of hemoglobin A, designated A2 and A3, were obtained by Kunkel (11), who used insoluble potato starch as a supporting medium for electrophoresis. Kunkel also was the first to measure quantitatively the amount of hemoglobin A2 and to correlate increases ...
متن کاملInheritable differences in the electrophoretic pattern of hemoglobin revealed in a local random-bred colony of albino rats.
I N THE COURSE OF OUR INVESTIGATION on the hemoglobins of various strains of laboratory rats with the use of starch-gel electrophoresis in a discontinuous buffer system, we have recently demonstrated the presence of three distinct patterns of hemoglobin, arbitrarily denoted as patterns I, II and III, in a local random-bred stock of Albino rats; out of 124 investigated male and female adult anim...
متن کاملHemoglobin electrophoresis at alkaline pH on agarose gels.
We describe a modified thin agarose-gel system for use in primary hemoglobin screening by electrophoresis. The system makes use of a Tris-EDTA-borate-buffered agarose gel (pH 8.8) and a sodium barbital electrophoresis buffer (pH 8.6). Separation is effected in 20 min at a constant potential of 250 V. Eight samples can be simultaneously separated and the patterns made visible in 47 min. There ar...
متن کاملThe abnormality of the primary structure of hemoglobin Shimonoseki.
Received April2, 1965 In 1960 Yamaoka et al. described one of the first well-documented abnormal hemoglobins to be found in Japan. This hemoglobin, designated Hb Shimonoseki for the city in which the family resides, constitutes approximately 20% of the hemoglobin of the heterozygotes and migrates with Hb S on paper electrophoresis at pH 8.6. The present communication concerns its further charac...
